Amakhosi, who left for Tunisia on Tuesday, take a two-goal advantage to this crucial second leg encounter as they aim to qualify for the final of the competition.
Chiefs coach Muhsin Ertugral expressed confidence that the Glamour Boys have what it takes to make history in African club competitions.
Ertugral, who had only 15 players in the first encounter, got a huge boost ahead of the return leg with news that both Thabo Mooki and Marco Mthembu would be ready to travel with the team.
Mooki, who scored both goals in the first leg, missed the league match against Ajax Cape Town and the Coca-Cola Cup quarterfinal clash against Manning Rangers with injury.
Mthembu has been out for several weeks with a lung infection but the medical staff have declared him fit for the Tunisia trip.
"This is good news for us," said coach Ertugral. "Mooki has always played a crucial role in the team and his return came at the right time. I am happy that Marco will also be available as his presence will give us balance at the back."
The coach also added that Doctor Khumalo, who inspired Chiefs to a 2-0 win over Africain in the first leg, would also be available for selection. "It's not going to be easy but we have to win this trophy for former president Nelson Mandela," said Ertugral.
Africain, who knocked out holders Zamalek 3-2 on aggregate in the quarterfinals, will have to score three goals without conceding to edge Amakhosi out.
In the other semifinal second leg to be played on Sunday, Inter Luanda of Angola take a healthy 3-0 lead to Kumbo Strikers of Cameroon, who had an easy start to the competition with a walkover win against Atletico on Burundi.
The Mandela Cup final will be played over two legs with the first leg taking place in two weeks and the second leg on the weekend of November 30-December 2.
